Gelsolin is a highly conserved, multifunctional actin-binding protein that severs, caps and nucleates actin filaments in response to Ca²⁺ and phosphoinositide signals, thereby orchestrating rapid cytoskeletal remodeling essential for cell motility, endocytosis, apoptosis and immune responses; it exists as both an 80–82 kDa cytoplasmic isoform and a slightly larger secreted plasma variant generated by alternative transcription initiation, each contributing to tissue repair and host defense by clearing actin filaments released from damaged cells, and its dysregulation—through mutation, proteolytic cleavage or altered expression—is implicated in cancer metastasis, neurodegeneration, amyloidosis and other human diseases.

IHC shows positive staining in paraffin-embedded human tonsil. Anti-Gelsolin antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
