USP1 (ubiquitin-specific protease 1) is a cysteine protease deubiquitinase that forms a tight complex with its activator UAF1 to remove monoubiquitin from key DNA-repair substrates—FANCD2/FANCI in the Fanconi-anemia pathway and PCNA during translesion-synthesis—thereby licensing accurate interstrand-crosslink repair, replication-fork stability, and genome integrity, while its frequent overexpression in cancers drives chemoresistance by also stabilizing oncogenic proteins such as ID1-3, EZH2, TAZ and survivin, making USP1 a promising therapeutic target.
