NEK7, a 34.5 kDa serine/threonine kinase of the NIMA-related family, is essential for mitotic entry, spindle organization and cytokinesis through phosphorylation of substrates such as EML4 and RPS6KB1, and is also indispensable for NLRP3 (and possibly NLRP1) inflammasome activation downstream of K⁺ efflux, where it binds NLRP3 via its catalytic domain—without requiring kinase activity—to promote oligomerization, ASC speck formation and caspase-1-dependent IL-1β/IL-18 release, thereby acting as a switch that mutually excludes mitosis from inflammasome signaling and links cell-cycle control to innate immunity and inflammatory diseases.

IHC shows positive staining in paraffin-embedded human colon. Anti-NEK7 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in Jurkat cells. Anti- NEK7 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
