GBP5 (Guanylate-Binding Protein 5) is a ~67 kDa interferon-inducible large GTPase that polymerizes on membranes and cytosolic surfaces to exert antimicrobial and inflammatory signaling functions; upon pathogen recognition it catalyzes GTP hydrolysis, oligomerizes into coat-like structures that destabilize bacterial or parasitophorous vacuole membranes, recruits the NLRP3 inflammasome via direct CARD-CARD interaction to drive caspase-1–dependent IL-1β/IL-18 maturation, and modulates autophagy and pyroptosis, thereby acting as a critical cytosolic pattern recognition receptor that links innate immunity to cell-autonomous defense against bacteria, viruses, and protozoa while its dysregulation contributes to autoinflammatory diseases and cancer.

ICC analysis of THP-1 cells treated with LPS (1μg/ml,24h) and hIFN-γ (40 ng/ml,24 h) (top panel) and untreated THP-1 cells (below panel). Anti-GBP5 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
