背景介绍

Protein Interacting with C Kinase 1 (PICK1) is a 416-residue peripheral membrane scaffolding protein encoded by the PICK1 gene, comprising an N-terminal PDZ domain that binds the C-termini of membrane proteins such as AMPA-receptor subunits GluA2/3, dopamine transporter and acid-sensing ion channels, a central BAR domain that senses and induces membrane curvature, and a C-terminal acidic region; through PDZ-mediated interactions it clusters and traffics glutamate receptors to and from synapses, thereby regulating excitatory synaptic strength and plasticity underlying learning and memory, while its BAR domain cooperates with actin and endocytic machinery to control clathrin-mediated endocytosis, synaptic-vesicle recycling and dense-core vesicle biogenesis, and disruption of PICK1 in mice leads to impaired long-term depression, male infertility and neurotransmission deficits .
