  • Autophagy-related protein 5 (ATG5), encoded by the ATG5 gene on chromosome 6, is a core autophagy protein that forms a complex with ATG12 and ATG16L1 (the Atg12-Atg5-Atg16 complex), which is essential for lipidation of LC3 (microtubule-associated protein 1 light chain 3B) and subsequent autophagosome elongation and maturation. Beyond autophagy, ATG5 also has non-autophagic roles, including promoting apoptosis via mitochondrial translocation and cytochrome c release, regulating immune responses (e.g., antigen presentation and lymphocyte development), and affecting hematopoietic stem cell maintenance. ATG5 deficiency (e.g., in knockout mice) is neonatal lethal, causing autophagy defects, neurodegeneration, and impaired fertility, while its overexpression extends lifespan. Additionally, ATG5 mutations are linked to human diseases (e.g., ataxia), and it plays a role in viral infections (e.g., HIV-1) by modulating host antiviral responses.
