Carbonic anhydrase IV (CA4) is a glycosylphosphatidylinositol-anchored membrane-bound zinc metalloenzyme that catalyzes the reversible hydration of CO₂ to bicarbonate and protons, playing a pivotal role in CO₂ and pH homeostasis across ocular, renal, pulmonary, and cerebrovascular endothelia; expressed abundantly on the luminal surface of capillaries and in the renal proximal tubule brush border, CA4 facilitates rapid CO₂ elimination, modulates cerebrovascular reactivity, and supports retinal photoreceptor function, with mutations linked to retinitis pigmentosa 17 and its pharmacological inhibition explored for glaucoma, altitude sickness, and edema therapy.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-CA4 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
