背景介绍

  • Cereblon (CRBN), encoded by the CRBN gene located on chromosome 3p26.2, is an evolutionarily conserved 442-amino-acid substrate receptor of the CRL4A E3 ubiquitin ligase complex, which ubiquitinates target proteins for proteasomal degradation, thereby influencing developmental processes (e.g., limb and auditory vesicle formation), regulating potassium channels, and modulating immune functions, including T-cell proliferation and activation. CRBN also serves as the primary molecular target for thalidomide and its immunomodulatory drug derivatives (lenalidomide, pomalidomide), which inhibit CRBN autoubiquitination and modulate ubiquitination of downstream substrates, thereby exerting anti-myeloma and teratogenic effects. Mutations or dysregulation of CRBN are linked to autosomal recessive nonsyndromic mental retardation and may affect cell metabolism and disease development.
