Prosaposin (PSAP) is a highly conserved, 68- to 73-kDa glycoprotein encoded by the PSAP gene on chromosome 10q22.1 that serves dual intracellular and extracellular roles: inside the endolysosomal system it is proteolytically cleaved into four small lysosomal activator proteins (saposins A–D) that bind either specific hydrolases or their glycosphingolipid substrates to enable sphingolipid catabolism, while outside the cell PSAP acts as a secreted neurotrophic factor that supports neuronal survival, neurite outgrowth and ERK signaling, can be endocytosed via LRP1 or CI-M6PR, and has recently been implicated in cancer progression, metastasis and as an emerging therapeutic target in tumors.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
