背景介绍

BCL-6 protein is a transcriptional repressor that plays a crucial role in the regulation of B cell differentiation and proliferation. It is primarily expressed in germinal center B cells and is essential for the formation and function of germinal centers, which are specialized microenvironments within lymphoid tissues where B cells undergo somatic hypermutation and class switch recombination to generate high-affinity antibodies. BCL-6 achieves its regulatory functions by binding to specific DNA sequences in the promoters of target genes, thereby inhibiting their transcription. Abnormal expression or function of BCL-6 is associated with various B cell lymphomas, making it an important biomarker and therapeutic target in the study and treatment of these malignancies.

- Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized Jurakt (Human T cell leukemia T lymphocyte, left) / Ramos (Human Burkitt's lymphoma B lymphocyte, right) labelling BCL-6 antibody at 1/50 dilution (1 μg) / (Red) compared with a Rabbit monoclonal IgG (Black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (Blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.
  Negative control: Jurkat

- IHC shows positive staining in paraffin-embedded human tonsil. Anti- BCL-6 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
