Stress-activated protein kinases (SAPK)/c-Jun N-terminal kinases (JNK) are members of the mitogen-activated protein kinase (MAPK) family, activated by various environmental stresses such as UV irradiation, inflammatory cytokines, and growth factors. They play crucial roles in regulating cell proliferation, apoptosis, motility, metabolism, DNA repair, and immune responses. The JNK/SAPK pathway involves a three-tiered kinase cascade, with MAPK kinases (MKKs) like MKK4 and MKK7 phosphorylating and activating JNKs. This pathway is essential for cellular adaptation to stress and has been implicated in diseases such as cancer and metabolic disorders.

- Flow cytometric analysis of 4% PFA fixed 90% methanol permeabilized NIH/3T3 (mouse embryonic fibroblast) labelling SAPK/JNK antibody at 1/50 dilution (1 μg)/ (red) compared with a Rabbit monoclonal IgG (black) isotype control and an unlabelled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat Anti - Rabbit IgG Alexa Fluor® 488 was used as the secondary antibody.

- ICC shows positive staining in NIH/3T3 cells. Anti- SAPK/JNK antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
