PRAME (Preferentially Expressed Antigen of Melanoma) is a protein encoded by the PRAME gene, belonging to the cancer-testis antigen family. It is predominantly expressed in various cancers, including melanoma, breast cancer, lung cancer, and leukemia, but is largely absent in normal tissues except for testis, ovary, placenta, and a few others. PRAME can inhibit retinoic acid signaling, thereby promoting cell proliferation and contributing to tumorigenesis. Due to its restricted expression pattern, PRAME is considered a promising target for cancer immunotherapy, with ongoing research exploring PRAME-specific vaccines and cellular immunotherapies.

免疫细胞化学
ICC shows positive staining in K562 cells. Anti- PRAME antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
