背景介绍
  • EGR1 protein, also known as Early Growth Response 1 or NGFI-A, is a mammalian transcription factor encoded by the EGR1 gene. It belongs to the EGR family of Cys2His2-type zinc finger proteins and acts as a nuclear protein regulating transcription. EGR1 plays a crucial role in various biological processes, including cell differentiation, proliferation, and apoptosis. It is involved in the regulation of multiple genes, some of which are tumor suppressors like TGFβ1, p53, and PTEN. However, its role in cancer is complex; while it is overexpressed in some cancers like colorectal and gastric cancer, leading to poor prognosis, it also acts as a tumor suppressor in others, such as rhabdomyosarcoma. Additionally, EGR1 is implicated in neuronal plasticity and cardiovascular diseases.

    • IHC shows positive staining in paraffin-embedded human kidney. Anti-EGR1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Anti-EGR1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin.   • ChIP

    • Chromatin immunoprecipitation (ChIP) was performed on HeLa+hTNF-α (20 ng/ml, 5 min) cells cross-linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactions used EGR1 Rabbit Polyclonal Antibody and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
      Post-immunoprecipitation, both samples were washed, eluted, and cross-links reversed. Purified DNA was analyzed by qPCR.
      qPCR showed the enrichment of ATAD2, EGR1 and SAT-α in EGR1 Rabbit
      Polyclonal Antibody-immunoprecipitated sample.
