IFT81 is a critical intracellular "cargo transporter" that serves as a core component of the intraflagellar transport (IFT) complex B. It primarily collaborates with IFT74 to form a specific tubulin-binding module, responsible for precisely transporting the tubulin required for cilium assembly to the designated sites. IFT81 binds to tubulin through its N-terminal calmodulin-homology (CH) domain, and another key function is its cooperative action with IFT74 to jointly accomplish the recognition and transport of tubulin. Therefore, IFT81 is essential for ciliogenesis. In addition, it plays important roles in multiple biological processes, including participation in the regulation of the Sonic Hedgehog signaling pathway and involvement in sperm flagellum assembly and elongation during spermatogenesis. Mutations in this gene have also been associated with short-rib polydactyly syndrome.
