背景介绍

  • Sonic Hedgehog (Shh) is a crucial secreted signaling protein and the most well-studied member of the Hedgehog family, playing an irreplaceable role in embryonic development. It acts as a morphogen to govern the dorsoventral patterning of the neural tube, the anteroposterior arrangement of limbs, and the formation of central nervous system structures. Upon binding to the Patched (Ptc) receptor on target cells, Shh relieves the inhibition of Smoothened (Smo), triggering an intracellular signaling cascade that activates Gli transcription factors to regulate specific gene expression. Beyond its developmental functions, Shh is essential for tissue maintenance and regeneration, and its dysregulated signaling is frequently implicated in various human cancers and severe congenital defects, such as holoprosencephaly.
