  • OGDH is the core subunit of the α-ketoglutarate dehydrogenase complex (OGDC) in the mitochondrial matrix. As one of the key rate-limiting enzymes of the tricarboxylic acid (TCA) cycle, it catalyzes the oxidative decarboxylation of α-ketoglutarate to succinyl-CoA and NADH, connecting two important metabolic nodes in the cycle and playing a critical role in cellular energy metabolism and redox balance. This enzyme is encoded by the OGDH gene, which is located on human chromosome 7 and encodes a precursor protein with an N-terminal mitochondrial targeting sequence that is processed to form the functional mature enzyme. Structurally, the OGDH protein consists of three domains: the N-terminal domain is responsible for binding the coenzyme thiamine pyrophosphate (TPP) to support substrate decarboxylation, the central region mediates homodimer formation, and the C-terminus contains the interaction interface for binding to dihydrolipoamide dehydrogenase (E3). Due to its central role in metabolism, OGDH activity is regulated by multiple mechanisms, including substrate availability, product inhibition, and phosphorylation modification. At the pathological level, OGDH dysfunction is closely associated with various neurological disorders; studies have found that its activity is significantly reduced in brain tissues of patients with Alzheimer's disease and Parkinson's disease, representing an important mechanism by which impaired energy metabolism contributes to neurodegeneration. In addition, OGDH gene mutations have also been linked to neurodevelopmental disorders such as early-onset dystonia and Leigh syndrome. In recent years, OGDH has also been recognized as a metabolic vulnerability in multiple tumors, and its inhibition represents a therapeutic strategy for targeting tumor metabolism.

    • ICC shows positive staining in HepG2 cells. Anti-OGDH antibody was used at 1/400 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Mouse IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
