HSP60 is a highly conserved and critically important molecular chaperone in cells. Its core function is to assist in the proper folding and assembly of nascent polypeptide chains, as well as the refolding of denatured proteins under stress conditions, thereby maintaining intracellular protein homeostasis. HSP60 typically exists as a double-ring oligomeric structure composed of 14 subunits (i.e., a "barrel-shaped" structure), and through ATP hydrolysis-driven conformational changes, it binds to the hydrophobic surfaces of substrate proteins to provide an isolated microenvironment that promotes the formation of correct conformations. In addition to its classical chaperone functions in the cytoplasm and mitochondria, HSP60 is also involved in the transmembrane transport of mitochondrial proteins, and accumulating evidence indicates that it can be secreted extracellularly or appear on the cell surface, exerting non-classical functions such as immunomodulation and induction of inflammatory signaling. HSP60 expression is significantly upregulated under stress conditions such as heat shock and oxidative stress. Its dysfunction is not only associated with various neurodegenerative diseases and heart diseases, but its expression levels are also markedly altered in multiple tumors, including breast cancer and colorectal cancer, closely correlating with malignant progression and drug resistance.
