  • FRS2-α (fibroblast growth factor receptor substrate 2-α) is a key adaptor protein characterized by an N-terminal PTB domain and multiple C-terminal tyrosine phosphorylation sites. It specifically anchors to the juxtamembrane regions of activated FGFR (fibroblast growth factor receptor) and TRK (tropomyosin receptor kinase) receptors. Upon phosphorylation by Src family kinases and others, it recruits the GRB2-SOS complex and activates the RAS-MAPK signaling pathway. Simultaneously, FRS2-α can independently initiate the PI3K-AKT pathway, thereby playing a core regulatory role in cell proliferation, differentiation, survival, and migration. Notably, FRS2-α not only serves as a "platform" for receptor signal amplification but also participates in signal negative feedback regulation: its downstream effector ERK can phosphorylate multiple serine/threonine sites on FRS2-α, inhibiting the binding of the PTB domain to the receptor, forming a classical negative feedback loop. In various tumors, amplification or overexpression of the FRS2-α gene leads to excessive activation of signaling pathways, promoting tumor progression, making it a potential therapeutic target. Furthermore, FRS2-α also plays important roles in metabolic regulation and nervous system development, and its mutations are associated with specific skeletal developmental abnormalities.
