  • YY1 is a ubiquitously expressed multifunctional zinc finger transcription factor belonging to the GLI-Krüppel family, named for its dual "yin-yang" regulatory property of both activating and repressing target gene transcription. The key structural feature of this protein is its C-terminal region containing four tandem C2H2-type zinc finger domains responsible for sequence-specific recognition and binding to the core DNA motif (5'-CCGCCATN-3'), while the N-terminal histidine/proline-rich region and acidic domain mediate interactions with various transcriptional cofactors (such as p300 and HDACs) and epigenetic regulatory proteins. YY1 plays a central regulatory network role in cell biology, participating in numerous physiological processes including embryonic development, cell proliferation, differentiation, apoptosis, DNA repair, and viral replication. Its multifunctionality is reflected in the following: on one hand, it represses target gene transcription by directly occupying promoter regions or recruiting histone deacetylases; on the other hand, it activates gene expression by recruiting histone acetyltransferases or cooperating with other transcription factors (such as Myc and p53). YY1 also plays an important architectural role, mediating the formation of DNA loops that bring distant regulatory elements (enhancers and promoters) into close proximity, thereby coordinating the expression of complex gene loci. Clinical studies have shown that YY1 is overexpressed in most tumors (such as lymphoma, breast cancer, prostate cancer, and pancreatic cancer), promoting tumor proliferation, invasion, metastasis, and chemoresistance by regulating epithelial-mesenchymal transition, glycolytic metabolic reprogramming, and anti-apoptotic genes. Conversely, in certain tumors, YY1 can also function as a tumor suppressor. Additionally, YY1 gene mutations are associated with a rare X-linked recessive disorder—Gabriele-de Vries syndrome, characterized by developmental delay, intellectual disability, and congenital abnormalities. YY1 is also involved in regulating pluripotent stem cell self-renewal and somatic cell reprogramming, serving as a key factor in maintaining embryonic stem cell identity.
