  • ALDOC (Aldolase C, also known as fructose-bisphosphate aldolase C) is a key isozyme in the glycolytic metabolic pathway, specifically and highly expressed in neurons of the central nervous system. As an important member of the class I aldolase family, this protein reversibly catalyzes the cleavage of fructose-1,6-bisphosphate into dihydroxyacetone phosphate and glyceraldehyde-3-phosphate—a reaction that plays a central role not only in glucose breakdown for energy production but also in gluconeogenesis. Distinguished from ALDOA, which is highly expressed in muscle, and ALDOB, which is highly expressed in the liver, ALDOC is predominantly enriched in brain tissue. Its functions extend beyond classical metabolic pathways, with growing evidence indicating that it also possesses non-glycolytic functions related to cytoskeletal dynamics regulation, synaptic vesicle trafficking, and neuronal differentiation. Clinical studies have shown that abnormal expression or dysfunction of ALDOC is closely associated with various neurological disorders, including malignant progression of glioma, metabolic dysregulation in Alzheimer's disease, and neuronal damage following cerebral ischemia, highlighting its value as a key regulator of metabolic balance and cellular homeostasis in the central nervous system, as well as a potential disease biomarker.
