CD138 (syndecan-1) is a type-I transmembrane heparan-sulfate/chondroitin-sulfate proteoglycan that tethers the cytoskeleton to the extracellular matrix, mediating adhesion, migration, and exosome biogenesis while acting as a co-receptor for growth factors and chemokines. It is constitutively expressed on epithelial cells and terminally differentiated plasma cells (making it a selective marker for plasma-cell neoplasms), but absent from mature B cells. Shed soluble CD138 is a prognostic indicator in multiple myeloma and other cancers; its overexpression or loss correlates with tumor progression, angiogenesis, and metastasis in solid tumors such as breast, colorectal, and liver cancers.

Flow cytometric analysis of Mouse CD138 expression on C57BL/6 mouse bone marrow. C57BL/6 mouse bone marrow was stained with APC Rat Anti-Mouse CD45R/B220 antibody and either Alexa Fluor® 488 Rat IgG2a, κ Isotype Control (Left panel) or SDT Alexa Fluor® 488 Mouse Anti- Human CD138 antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
