Granzyme B is a 32-kDa serine protease stored in the cytolytic granules of cytotoxic T lymphocytes and natural killer cells, whose immune function centers on perforin-assisted translocation into target cells where it directly cleaves and activates pro-apoptotic substrates such as procaspase-3, Bid and PARP, thereby igniting both caspase-dependent and caspase-independent mitochondrial apoptosis pathways to eliminate virus-infected or malignant cells.
流式分析
Flow cytometric analysis of 4% PFA fixed 0.1% Tween-20 permeabilized PBMC (Human peripheral blood mononuclear cells) labelling Granzyme B antibody at 1/200 dilution (1 μg)/ (Right panel) compared with a Mouse IgG1, κ Isotype Control (Left panel). Goat Anti - Mouse IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD8 - Brilliant Violet 421™ antibody separately.
