CD169 (also called Siglec-1 or sialoadhesin) is a 175 kDa type I transmembrane glycoprotein of the immunoglobulin superfamily expressed predominantly by subsets of tissue-resident macrophages and some dendritic cells; it contains 17 extracellular Ig-like domains whose distal V-set domain binds α2,3-linked sialic acids on host glycoproteins and glycolipids or on pathogens, enabling cell–cell and cell–matrix adhesion during inflammation, pathogen capture (e.g., PRRSV, SARS-CoV-2), antigen presentation, and modulation of innate and adaptive immunity, while its expression is up-regulated by interferons and correlates with disease activity in infections, inflammatory disorders, and cancer where CD169⁺ macrophages in tumor-draining lymph nodes are associated with better prognosis.
流式分析
Flow cytometric analysis of peripheral blood monocyte-derived dendritic cells labelling Mouse CD169 antibody at 1/200 dilution (1 μg) / (Right panel) compared with a Mouse IgG1, κ Isotype Control / (Left panel). Goat Anti-Mouse IgG Alexa Fluor® 647 was used as the secondary antibody. Then cells were stained with CD11c - FITC Antibody separately.
