Surface Antigen 1 (SAG1) is a major surface protein of Toxoplasma gondii and one of the most valuable biomarkers for toxoplasmosis diagnosis in humans and animals. Highly conserved across different Toxoplasma strains, SAG1 is abundantly expressed on tachyzoites, the invasive stage of the parasite, enabling early and sensitive detection of acute infection. In serological tests, SAG1 effectively induces strong specific antibody responses in infected hosts. It is widely applied in ELISA, lateral flow assays and immunofluorescence assays to screen anti-Toxoplasma IgG and IgM, distinguishing acute, chronic and past infections. For livestock, SAG1-based detection helps reduce vertical transmission, abortion risks and economic losses in animal husbandry. In human clinical settings, it is critical for high-risk groups such as pregnant women and immunocompromised patients, facilitating timely intervention to prevent congenital toxoplasmosis and life-threatening complications. With high specificity and immunogenicity, SAG1 outperforms many other antigens. It serves not only as a reliable diagnostic target but also provides a solid foundation for epidemiological surveillance and vaccine research against toxoplasmosis.
