- Tissue factor (TF) is a lipoprotein cofactor that markedly enhances the proteolytic activation of factors IX and X by factor VIIa. The functional activity of TF is inhibited by serum in a time- and temperature-dependent fashion. It is a transmembrane protein that is consitutively expressed in subendothelial cells throughout the vasculature and is inducible on endothelial cells and monocytes. Damage to the vessel wall exposes Coagulation Factor III to circulating Factor VII, triggering the proteolytic cascade that results in fibrin clot formation. Coagulation Factor III plays an important role in thrombotic diseases, disseminated intravascular coagulation, and atherosclerosis. It also plays a role in non-hemostatic inflammation, angiogenesis, and tumor progression. Many studies have shown that levels of circulating TF in the form of MPs are increased in various diseases, including cardiovascular disease, sepsis, diabetes, and cancer. This MP TF likely contributes to the increased incidence of thrombosis observed in these diseases. One study showed that TF-positive MPs isolated from the pericardial blood of cardiac surgery patients increased venous thrombosis in the rat model.
