BSP II (Bone Sialoprotein II) is a heavily sialylated, secreted glycoprotein that constitutes up to 10 % of bone’s non-collagenous matrix, where its clusters of acidic amino acids confer high-affinity binding to hydroxyapatite, its poly-Glu sequences nucleate mineral deposition, and its C-terminal RGD motif supports osteoblastic and odontoblastic adhesion via αvβ3 integrin, thereby acting as a key structural and signaling scaffold that synchronizes the onset of matrix mineralization with cell attachment during skeletal and cementum formation.

IHC shows positive staining in paraffin-embedded human osteosarcoma. Anti-BSP II antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
