The SS18-SSX fusion protein is the defining oncogenic driver of synovial sarcoma, arising from the chromosomal translocation t(X;18)(p11;q11) that fuses the SS18 gene on chromosome 18 with SSX1, SSX2, or rarely SSX4 on the X chromosome, producing a chimeric protein in which the C-terminal 8 amino acids of SS18 are replaced by 78 residues from SSX; this fusion retains the N-terminal SNH domain of SS18 that mediates incorporation into SWI/SNF (BAF) chromatin-remodeling complexes, but the SSX moiety recruits polycomb repressive complexes (especially PRC1.1 via KDM2B) and binds H2AK119ub-rich regions, thereby hijacking BAF targeting to aberrantly activate developmentally repressed genes, evict the tumor-suppressive BAF47 (SMARCB1/INI1) subunit, and globally reprogram the epigenetic landscape to enforce malignant transformation.

IHC shows positive staining in paraffin-embedded human synovial sarcoma. Anti- SS18-SSX antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows negative staining in paraffin-embedded human colon. Anti- SS18-SSX antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use).
