Synaptopodin is an actin-associated protein concentrated in ~30 % of large dendritic spines of telencephalic neurons where it nucleates the spine apparatus—stacks of smooth endoplasmic reticulum that function as local calcium stores and sites of protein synthesis—thereby enabling PKA- and activity-dependent enlargement of spine heads, trafficking of GluA1-containing AMPA receptors, and the expression of long-term potentiation and memory formation, so that synaptopodin-deficient mice lack spine apparatuses, show blunted LTP, and exhibit impaired spatial learning.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-Synaptopodin antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
