背景介绍

  • AKAP4, the most abundant protein of the sperm fibrous sheath, is a testis-enriched scaffolding protein that simultaneously binds PKA via dual-specificity anchoring sites and PKC through polybasic motifs, thereby acting as a cAMP/PKA ↔ PKC/ERK1/2 switch that is phosphorylated on Ser/Tyr during capacitation to direct the kinase cascade to the flagellar principal piece, mid-piece and post-acrosomal region, enabling progressive motility and male fertility while also moonlighting in multiple cancers where its cytoplasmic/membrane accumulation drives proliferation, EMT and metastasis through the Shh and Wnt/β-catenin pathways.

    • IHC shows positive staining in paraffin-embedded human testis. Anti-AKAP4 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

    • Negative control: IHC shows negative staining in paraffin-embedded human colon. Anti-AKAP4 antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
