SLC6A1 encodes GAT-1, the principal Na⁺-coupled transporter that clears the inhibitory neurotransmitter GABA from the synaptic cleft by pumping it back into neurons and astrocytes, thereby terminating GABAergic signaling and maintaining excitatory–inhibitory balance throughout the CNS; mutations that misfold GAT-1 or abolish its plasma-membrane trafficking reduce synaptic GABA clearance, causing epilepsy, intellectual disability, autism spectrum disorders, ADHD, and mood dysfunction, whereas chemical or pharmacological chaperones can rescue some variants by restoring correct folding and surface expression.

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-SLC6A1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
