Stromal cell-derived factor-1 (SDF-1), also designated CXCL12, is a 67-amino-acid constitutively secreted chemokine that signals exclusively through the Gαi-protein-coupled receptor CXCR4 (and atypical CXCR7) to orchestrate embryonic development, adult stem-cell homing, immune surveillance, vascular remodeling, and neural circuitry; its 3-stranded β-sheet flanked by an α-helix and disulfide-stabilized loops binds CXCR4 with nanomolar affinity, triggering PI3K–Akt, MAPK/ERK, and calcium flux that mediate chemotaxis, survival, and proliferation, whereas dysregulated SDF-1/CXCR4 signaling fuels cancer metastasis, inflammatory disease, and HIV entry, making the pathway a prime target for small-molecule antagonists, peptide analogs, and neutralizing antibodies in oncology and regenerative medicine.

IHC shows positive staining in paraffin-embedded human ×××. Anti-SDF1/CXCL12 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
