Myogenin is a basic helix-loop-helix transcription factor encoded by the MYOG gene that serves as the master regulator of skeletal muscle differentiation, binding to E-box motifs in promoters of muscle-specific genes like desmin, myosin heavy chain, and the acetylcholine receptor to orchestrate terminal differentiation and fusion of myoblasts into multinucleated myotubes; its expression is induced by the upstream myogenic regulatory factors MyoD and Myf5, and its activity is modulated by interactions with E proteins, p300/CBP coactivators, and MEF2 family members, while in pathophysiology MYOG mutations or altered expression are linked to congenital myopathies and its ectopic expression in tumors is exploited as a sensitive immunohistochemical marker for rhabdomyosarcoma.

IHC shows positive staining in paraffin-embedded human rhabdomyosarcoma. Anti-Myogenin antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
