Orthopedia homeobox protein (OTP) is a nuclear transcription factor encoded on chromosome 5q14.1 that belongs to the homeobox family and is indispensable for embryonic neurodevelopment ; in clinical oncology it serves as a highly sensitive and specific immunohistochemical marker that strongly stains typical pulmonary carcinoids while remaining negative in most non-lung well-differentiated neuroendocrine tumors, with negative or weak nuclear OTP expression correlating with worse prognosis in carcinoid patients .

Negative control: IHC shows negative staining in paraffin-embedded human small cell lung cancer. Anti-OTP antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
