The regenerating gene (REG) family members, REG1∼4, are small secreted lectin-like proteins involved in hepatic, pancreatic, gastric and intestinal cell proliferation and differentiation. It has been reported that aberrant expression of REG4 is associated with the growth, survival, adhesion and resistance to apoptosis of tumor cells. Furthermore, REG4 has been identified as one of the genes up-regulated in gastric cancer-initiating cells and is thus used as a novel marker for these cells.
免疫组化
IHC shows positive staining in paraffin-embedded human colon. Anti- Reg4 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows negative staining in paraffin-embedded human colon. Anti- Reg4 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 555) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
