Zinc fingerprint structure transcription factor, responsible for regulating the self -update of embryo stem cells, is a key gene in many tumors. It plays an important role in embryo development. It is stem cell marker and tumor embryo protein, similar to AFP. SALL4 is the sensitivity and specific marker of seminoma and ovarian germ cell tumor. In pathology, the diagnosis of germ cell tumors is mainly used. It is also used in the identification of gastric liver cancer and Hepatocellular carcinoma, and diagnosis of yolk sac tumor and clear cell carcinoma of the ovary.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ICC shows positive staining in NCCIT cells (top panel) and negative staining in HeLa cells (below panel). Anti- SALL4 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
