TFE3 is a member of the MiT family of the bHLH-leucine zipper transcription factor. Fusions of the TFE3 gene with several different partner genes occur in several tumours, resulting in nuclear expression of the TFE3 protein. Research studies indicate that TFE3 and other family members play roles in development, organelle biogenesis, nutrient sensing, autophagy, and energy metabolism.

IHC shows positive staining in paraffin-embedded human kidney. Anti-TFE3 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

ChIP
Chromatin immunoprecipitation (ChIP) was performed on HeLa cells cross - linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactions used TFE3 Recombinant Rabbit mAb (SDT-760-34) and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
Post-immunoprecipitation, both samples were washed, eluted, and cross-links reversed. Purified DNA was analyzed by qPCR.
qPCR showed the enrichment of GLA, ATP6VIH and SAT-α in TFE3 Recombinant
Rabbit mAb (SDT-760-34)-immunoprecipitated sample.
