背景介绍
  • Phospho-RIPK1 (Ser166) is a phosphorylated form of Receptor-Interacting Serine/Threonine-Protein Kinase 1 (RIPK1), a key regulator of TNF-mediated apoptosis, necroptosis, and inflammatory pathways. This protein consists of an N-terminal kinase domain, an intermediate domain containing the RIP homotypic interaction motif (RHIM), and a C-terminal death domain. When phosphorylated at Ser166, RIPK1 is involved in the formation of necrosis-inducing complexes and is considered a biomarker for the activation of RIPK1 kinase and necroptosis. This phosphorylation can be effectively inhibited by Nec-1, a necroptosis inhibitor.

检测磷酸化蛋白时,推荐有什么封闭体系?

我们推荐客户使用TPST+5%脱脂奶粉来稀释一抗,进行封闭。 虽然BSA被推荐为WB检测磷酸化蛋白的常用封闭剂,但是脱脂奶粉获取更加方便,覆盖更广泛的非特异性结合位点,在一抗性能优越的前提下,使用脱脂奶粉封闭性价比更高
