背景介绍

  • SMARCD1, also known as BAF60A, is a protein that is part of the SWI/SNF family of chromatin remodeling complexes. It plays a crucial role in regulating gene transcription by altering chromatin structure, which involves the alteration of DNA-nucleosome topology in an ATP-dependent manner. SMARCD1 is involved in various biological processes, including neurogenesis and the regulation of lipid metabolism. It is also implicated in diseases such as Coffin-Siris Syndrome and has been identified as a prognostic marker in liver hepatocellular carcinoma. The protein is ubiquitously expressed in the nucleus of various tissues and is essential for the activation of specific gene signatures in multipotent progenitors.

    • IHC shows positive staining in paraffin-embedded human testis. Anti-SMARCD1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

    • ICC shows positive staining in NIH/3T3 cells. Anti- SMARCD1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C.
