背景介绍
  • Histone H4 (acetyl K20), or H4K20ac, is a post-translational modification that is generally associated with gene repression in human cells. Contrary to most histone acetylation events that are linked to gene activation, H4K20ac is enriched around the transcription start sites (TSSs) of minimally expressed genes and within the gene body of expressed genes. This unique acetylation mark shows little correlation with known histone modifications, including histone H3 methylations. Transcriptional activators are mostly excluded from genes enriched with H4K20ac, while the transcription repressor NRSF/REST co-localizes with this mark, suggesting that H4K20ac plays a distinct role in the epigenetic regulation of gene expression.

    • ICC analysis of HeLa cells treated with Trichostatin A (1 μM, 18 hr) (top panel) and HeLa cells untreated with Trichostatin A (below panel). Anti- Histone H4 (acetyl K20) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

  • ChIP

    • Chromatin immunoprecipitation (ChIP) was performed on HeLa + Sodium butyrate (30mM, 4hr) cells cross - linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactions used Histone H4 (acetyl K20) Recombinant Rabbit mAb (S-846-491) and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
      Post - immunoprecipitation, both samples were washed, eluted, and cross - links reversed. Purified DNA was analyzed by qPCR.

      qPCR showed the enrichment of CAMK2N1, FAM43B and MUL1 in Histone H4
      (acetyl K20) Recombinant Rabbit mAb (S-846-491)-immunoprecipitated sample.
