KDM5D, also known as JARID1D or SMCY, is a gene located on the Y chromosome that encodes a protein with zinc finger domains. This protein acts as a histone demethylase, specifically targeting H3K4me2/3 marks, which are associated with transcriptional activation. KDM5D plays a role in various biological processes, including the development and function of the male reproductive system. It has been implicated in the regulation of genes that control cell adhesion and immune response in cancer, with its upregulation potentially contributing to cancer progression and gender differences in tumor behavior. KDM5D is also associated with minor histocompatibility antigens, which can lead to graft rejection in transplant settings. Its dysfunction has been linked to spermatogenic failure, highlighting its importance in male fertility.

IHC shows positive staining in paraffin-embedded human colon. Anti-KDM5D antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
