背景介绍
  • Histone H4 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H4 is involved with the structure of the nucleosome of the 'beads on a string' organization. Histone H4 is an important protein in the structure and function of chromatin, where its sequence variants and variable modification states are thought to play a role in the dynamic and long term regulation of genes.

Anti-Propionyl-Histone H4 (Lys5) antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (red).

  • ChIP

    • Chromatin immunoprecipitation (ChIP) was performed on HeLa+Sodium butyrate (30mM, 4hr) cells cross - linked with 1% formaldehyde for 10 min, then chromatin was fragmented by sonication. Parallel reactions used Propionyl Histone H4 (Lys5) Recombinant Rabbit mAb (S-R093) and Rabbit mAb IgG Isotype Control (SDT-R173) at 1:50 for immunoprecipitation.
      Post - immunoprecipitation, both samples were washed, eluted, and cross - links reversed. Purified DNA was analyzed by qPCR.
      qPCR showed the enrichment of HBB and SAT-α in Propionyl-
      Histone H4 (Lys5) Recombinant Rabbit mAb (S-R093)-immunoprecipitated
      sample.
