Beta-secretase 2 (BACE2) is an aspartic protease encoded by a gene located on chromosome 21. It is expressed in various peripheral tissues and, to a lesser extent, in the central nervous system. Unlike its role in amyloid precursor protein (APP) processing, BACE2 primarily cleaves APP within the amyloid-beta domain, an action that may limit amyloid-beta peptide production. Its physiological functions extend to processes such as pancreatic beta-cell maintenance, skin pigmentation regulation, and skeletal muscle homeostasis. Research into BACE2 is also significant in the context of Down syndrome, due to the gene dosage effect associated with its chromosomal location.
