RNASE2, commonly known as Eosinophil-Derived Neurotoxin (EDN), is a secretory protein belonging to the RNase A superfamily. It is abundantly expressed in and secreted by eosinophils, a type of white blood cell. While it exhibits ribonuclease activity, its primary biological significance lies in its potent anti-pathogen and immunomodulatory functions. RNASE2 displays broad antiviral activity against single-stranded RNA viruses and possesses neurotoxic properties in vitro. It acts as an endogenous ligand for Toll-like receptor 2 (TLR2), thereby promoting dendritic cell-mediated adaptive Th2 immune responses. Its levels are clinically significant as a biomarker for eosinophil activation in diseases such as asthma, allergies, and helminth infections.
