Nicotinamide N-methyltransferase (NNMT) is a cytoplasmic enzyme that catalyzes the methylation of nicotinamide (vitamin B3) using S-adenosyl methionine (SAM) as the methyl donor, producing S-adenosylhomocysteine (SAH) and 1-methylnicotinamide (MNA). Its overexpression is implicated in various cancers, metabolic disorders, and aging, as it consumes SAM and influences global epigenetic patterns by altering the SAM:SAH ratio. NNMT is now recognized as a key metabolic enzyme with potential as a therapeutic target for cancer and obesity.
