背景介绍

  • Human TL1A (also known as TNFSF15) is a cytokine belonging to the tumor necrosis factor (TNF) superfamily, primarily expressed by vascular endothelial cells, macrophages, and dendritic cells. It functions by binding to its cognate receptor DR3 (Death Receptor 3) on activated T cells and innate lymphoid cells, delivering a potent co-stimulatory signal that strongly enhances the production of interferon-gamma and other inflammatory cytokines. This signaling axis plays a crucial role in amplifying and sustaining immune responses, particularly in mucosal tissues such as the intestines. Genetic studies have firmly established that variants in the TNFSF15 gene are significant risk factors for the development of chronic inflammatory bowel diseases, including Crohn's disease and ulcerative colitis. Beyond gut inflammation, TL1A is implicated in the pathogenesis of other autoimmune and inflammatory conditions, such as rheumatoid arthritis and psoriasis. Given its central role in driving pathogenic inflammation, the TL1A/DR3 pathway has emerged as a highly promising therapeutic target, with several neutralizing antibodies and inhibitors currently under clinical investigation.

      The concentrations of Human TL1A/TNFSF15 were measured and interpolated from the target standard curves and corrected for sample dilution.
      Sample is undiluted samples are as follows: HT-29 Cell culture supernatant was 2-fold dilution (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 60.36 pg/mL in HT-29 Cell culture supernatant.
      Sample is undiluted samples are as follows: HL-60 cell culture supernatant treated with 200nM PMA for 24h (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 96.39 pg/mL in PMA-treated HL-60 cell culture supernatant.
      Sample is undiluted samples are as follows: untreated HL-60 cell culture supernatant (50%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be <7.81 pg/mL in untreated HL-60 cell culture supernatant.

    • To assess the linearity of the assay, three samples were spiked with high concentrations of Human TL1A/TNFSF15 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
