Tri-Methyl Lysine (Kme3) is a post-translational modification (PTM) where three methyl groups (–CH₃) are added to the ε-amino group of a lysine residue, forming Nε-trimethyllysine. This modification is commonly found on histone proteins (e.g., H3K4me3, H3K9me3, H3K27me3) and plays a crucial role in regulating chromatin structure and gene expression by recruiting effector proteins such as chromatin remodelers and transcription factors. Trimethylation is catalyzed by lysine methyltransferases (KMTs, e.g., EZH2 for H3K27me3) and can be reversed by lysine demethylases (KDMs, e.g., JMJD2 family). Unlike mono- or di-methylation, trimethylation often marks stable epigenetic silencing (e.g., H3K27me3 in Polycomb repression) or active transcription (e.g., H3K4me3 at promoters). It is detected using antibody-based methods (ChIP-seq, IF, WB) or mass spectrometry, and its dysregulation is linked to cancer and developmental disorders.
